Using external mic on Mic/Aux versus independent input source

Is there any major difference between choosing the external mic as an audio source while disabling Mic/Aux from Settings or selecting the external mic from Mic/Aux? Thank you.

Audio in Settings is global, always ready, audio added per scene working only if scene is on PGM.

But no difference in say delay/lag between using either? Also what is PGM?

PGM - program, that screen and audio send
PRV - preview
Some people say: diffrences exists but but i can hear.
