"Enable Preview" is greyed out.

All of a sudden I see "Enable Preview" in the preview window is greyed out. This has become very difficult for me because I am unable to crop/scale/edit in the preview as it is directly showing in the program area. Please refer the attached screen shot. Someone please help!!! Thanks in advance.

Disabling preview in studio mode doesn't make sense, so its grayed out. In single window mode it can be used to disable the preview at all.

What exactly do you mean with "the preview as it is directly showing in the program area".
Whats a program area? Do you mean an fullscreen or windowed projector?

I believe the issue is you're stuck in Studio Mode. Click Studio Mode so it is no longer highlighted under your Controls and it should take you back to just one preview and you'll be able to crop/scale/edit the preview.
If your 'Control' Dock isn't already added to your OBS Dock, click Docks on the top left corner of OBS, click Controls so that a check mark appears next to it. Once that's added to your OBS, click Studio Mode to get back to just one preview and be able to edit/crop/scale the preview.
